Loopy has looked into it, basically said it would be very difficult due to the size. N3DS is packed very tightly without much available space.
I don't think he said it's impossible, but I think he's not actively doing R&D on it.

You need to plug it into a Line-In port (the port is typically coloured blue. mic is red, speakers are green etc). I do not think an all-in-one PC would have the port. As far as I know, there are no hardware changes between the Japanese and North American models. The differences are within the software, so it should not affect the capture card's compatibility with Japanese consoles.
